Metnase Mediates Resistance to Topoisomerase II Inhibitors in Breast Cancer Cells

Metnase levels predict breast cancer cell sensitivity to Topo IIα inhibitors.

Cells stably transfected with vector control, or Metnase shRNA were treated with VP-16 (A) or adriamycin (B) and colony survival was measured as described. Values are averages ±SEM for three determinations. (C) Cells transfected with control (shGFP) or Metnase shRNA were treated with 1 µM adriamycin for 24 hrs and apoptosis (annexin-V) and cell death (propidium iodide) were analyzed by FACS. Values are averages (+SEM) for three determinations.
