The effect of type I IFNs to SOCS2 mRNA induction in human moDCs.
moDCs were exposed to LPS (A) for indicated time periods for mRNA measurement of IFNα, IFNβ, IFNγ and IFNλ1 or incubated with IFNα (Bi), IFNβ (Bii), IFNγ (Biii) and IFNλ1 (Biv) for indicated time periods for SOCS2 mRNA measurement by qRT-PCR. (C) moDCs were pretreated for 30 min with or without 30 ug/ml neutralizing anti-IFNR2 antibodies or IgG2a isotype control mAb and then incubated for 4 h with or without LPS for SOCS2 mRNA measurement by qRT-PCR. Data shown are representative of three independent experiments and expressed as the –fold induction of the gene of interest at different time points compared to time point 0 where the values were set to 1(A–B) or different treatment conditions compared with medium that values were set to 1.
